ROC curve using strategy A (average value of wells must exceed threshold, black line) and strategy B (values in both wells must exceed threshold, dashed line) with thresholds ranging from 50 to 500 copies/ml. The top left point on the dashed line indicates the optimal threshold of 140 copies/ml corresponding to a sensitivity of 97.2% and specificity of 99.6%. We chose to use 150 copies because it matches that used for Taqman and the performance is nearly identical: sensitivity of 96.7% and specificity of 99.6%.
