Rescue Strategies for Protein Production in E. coli



E. coli is an attractive host for protein production because it is well-understood, fast-growing, inexpensive and facile to manipulate.Unfortunately, many proteins do not express to high levels or in properly folded form in this simple bacterial system.It may be necessary to utilize a different expression system in order to obtain full-length, biologically active proteins; however, there are many methods available to increase the likelihood of producing satisfactory levels of useful protein in E. coli. New vectors and host strains become available every year and novel methods appear in the literature on a frequent basis.An overview of rescue strategies for coercing misbehaved proteins to express in suitable yield in soluble form when produced in E. coli will be presented.
