All Rights Reserved.): Three-dimensional micro-computed tomography (μCT) reconstruction images of a femur from a 2-month-old mutant male lacking β-catenin, the central mediator of canonical Wnt signaling, selectively in osteocytes, terminally differentiated cells of the mesenchymal osteoblast lineage embedded within the mineralized bone matrix. Genetic ablation of β-catenin in osteocytes results in a dramatic low-bone-mass phenotype characterized by the absence of nearly all cancellous bone structures and distinct perforations in the long bone cortical shell, as clearly visible on these exemplary μCT images. High-resolution bone imaging and image processing were performed on a vivaCT40 (Scanco Medical). (See related article on p. 3071.)
