Detection of human iNKT by immunohistology

6B11, positive and isotype controls were used to stain frozen sections, since no specific 6B11 staining was observed on paraffin slides (not shown). Double immunofluorescence staining and laser confocal images of bronchial biopsies from a patient with severe asthma was performed as described in Material and Methods. Briefly, the frozen sections were stained with PE-conjugated CD4 (red), and FITC-conjugated 6B11 mAb (blue) and analyzed with confocal laser scanning microscopy.

C. The overlay results indicated that a significant number of the CD4+ infiltrating lymphocytes in asthma co-expressed the invariant TCR Vα24Jα18 (× 600).
