All Rights Reserved.): Confocal micrograph showing the calcium-induced anchoring of the transcription factor URE3-BP to the plasma membrane of an Entamoeba histolytica trophozoite mediated by the phospholipid-binding protein EhC2A. E. histolytica strain HM1:IMSS trophozoites were incubated under conditions that elevated intracellular Ca2+ and were fixed, permeabilized, and fluoroimmunostained for EhC2A (green) and URE3-BP (red), and nuclei were stained with DAPI (4′,6-diamidino-2-phenylindole) (blue). For trophozoites depleted of intracellular Ca2+, EhC2A was homogenously distributed throughout the amebic cytoplasm and URE3-BP was distributed throughout amebic nuclei and cytoplasm (not shown). The authors propose a novel mechanism of Ca2+-mediated transcriptional regulation via the sequestration of the transcription factor to the plasma membrane. (See related article on page 695.)
