Cover photograph (Copyright © 2010, American Society for Microbiology. All Rights Reserved.): Aberrant activation of the atypical Rho GTPase Wrch-1 disrupts epithelial morphogenesis. Expression of constitutively active Wrch-1(Q107L) or short hairpin RNA (shRNA) directed against Wrch-1 perturbs cystogenesis of MDCKII cells grown in three-dimensional culture, compared to scrambled shRNA and vector control cells. Morphogenesis of cells expressing the empty vector (top right) or Wrch-1(Q107L) (bottom right) was assessed by ZO-1 and E-cadherin staining. The actin cytoskeleton was visualized in cysts formed by cells expressing scrambled shRNA (top left) or Wrch-1 shRNA (bottom left) by phalloidin staining. These data highlight the importance of tight regulation of Wrch-1 activity for proper formation of epithelial cell architecture. (See related article in February 2009, vol. 29, no. 4, p. 1035.)
