Progressive inhibition of tubulin assembly with MAPs and paclitaxel by increasing concentrations of GDP. Each 200 μL reaction mixture contained tubulin at 1.0 mg mL-1 (10 μM), 0.75 mg mL-1 heat-treated MAPs, 10 μM paclitaxel, 0.1 M Mes (pH 6.9), and 1% (v/v) dimethyl sulfoxide (the drug solvent). GDP concentrations were as follows: curve 1, none; curve 2, 0.25 mM; curve 3, 0.5 mM; curve 4, 1.0 mM; curve 5, 2.0 mM; curve 6, 5.0 mM.
